DID your pleco die? He was probably eating the diatoms before you saw them. No it should not hurt your fish at moderate levels at least.
What fish do you have in their now?

DO you mean a red bleeding spot? or a raised red bump?
Do you know what your water parameters are?

Was the spot there before it died? Usually a sign of bacterial infection from poor water quality.

If the spot was there before it died, yes it got sick.
If the spot was after, maybe a fish took a bite out of it? Your pleco maybe?
